After this year's success, the Sea Eagles will return to face the Canberra Raiders on April 2 in round four.
In front of a crowd of 6,380, the maroon and white contingent posted a 36-0 victory against the Gold Coast Titans on April 17, 2021 in Mudgee.
Mid-Western Regional Council general manager, Brad Cam said given the number of Manly's supporters, the game is going to be "hugely successful".
"We're thrilled to welcome back the Sea Eagles for the second time after their hugely successful game at Glen Willow back in 2021," Mr Cam said.
"The club were particularly proactive with engaging the community and the region's youth last year and no doubt there will be a lot of excited fans pleased to see their return.
"NRL premiership games are of huge economic benefit to our region generating more than $1 million in local spending per game, and Council has worked hard to foster relationships within the NRL since the first Glen Willow game back in 2011.
"Since then Glen Willow has hosted four NRL premiership games and we're hoping more clubs will see the benefits of playing at the best regional stadium in NSW."
Ticketing details for the April game will be announced in the New Year.
