Members of the public reported the Eastern Long-Necked Turtles on the side of the Avon River, Gippsland, a kilometre north of Stratford, last week.
Wildlife investigators have inspected the area and are calling for the owner of a 4WD to come forward, after it appears the vehicle became bogged beside the river near the turtles.
"We'd like the owner of the 4WD to come forward and speak to us, so that we can eliminate them from our inquires," the Department of Environment said in a statement.
Anyone who has seen anything suspicious or has information about the 4WD owner should contact Crime Stoppers on 1800 333 000.
