Perry claimed 2-9 as the Hurricanes were restricted to 9-125 batting first at Blundstone Arena on Sunday.
She then combined with Indian teenager Shefali Verma to help steer the Sixers to victory with three balls to spare.
Perry made 27 before she was stumped off the bowling of Molly Strano to end a 63-run stand for the third wicket with Verma, after the early dismissals of Australia internationals Alyssa Healy (3) and Ashleigh Gardner (3).
Verma top-scored for the Sixers with her first half-century in WBBL cricket, the 17-year-old opener dismissed for 57 in the penultimate over.
Strano, the all-time leading wicket-taker in WBBL history, was the pick of Hobart's bowlers with 3-19 off her four overs including the wickets of Perry and Verma.
Earlier, the Hurricanes were sent in by Sixers' captain Perry and found themselves in early trouble slumping to 3-26 off six overs including a sensational one-handed catch by Verma's compatriot Radha Yadav to dismiss Mignon du Preez (1).
As well as Perry, Yadav (2-31), Nicole Bolton (2-29) and Maitlan Brown (2-23) all provided strong contributions with the ball for the Sixers.
Another India international, 18-year-old Richa Ghosh, was Hobart's best with the bat with a run-a-ball 46.
Defeat means the Hurricanes have lost their first two matches of the competition, after a six-wicket loss to the Melbourne Renegades on Saturday.
The Hurricanes-Sixers match was brought forward to be played in the morning on Sunday due to the delayed arrival into locked-down Hobart of the Perth Scorchers.
That decision allows Perth's clash with Brisbane Heat to now be played from 1.40pm AEDT, in what is now the final match of Sunday's double-header.
